  • Ah-rsceptor dependent changes of cytochrome P4501A1 (CYP4501A1/2) in natural- and induced status by a 3-methylcholanethrene (3-MC) and 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD) at FGS/NgaKist Mice with glomerulosclerosis were studied using immunohistochemical study. All experimental groups were showing the degree of high positive proteinuria and glomerulosclerosis in the kidney. Mice received dose of 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD), 40 ㎍/㎏ and 3-methylcholanethrene (3-MC), 80㎎/㎏ by oral administration, in the immunohistochemistry, a clear heterogeneous induction of cytochrome P4501A1/2 from uninduced- and induced mice was detected predominantly in periportal hepatocytes. These regional localization and expression indicate that not only merely dependent on exogenous factors in induced hepatocytes, but also endogenous factors in normal status. These findings provide new insight into the mechanism of Ah-receptor mediated induction by endogenous factors in the renalpathy. (rdf:langString) (en)
  • 사구제 경변증을 보이는 FGS/NgaKist mice에서 Ah receptor 조절물질로 알려진 3-methylcholanthrene (3-MC)및 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD)를 투여한 군과 무처치군에서 약물대사 효소인 사이토크롬 P4501A1/2단백질 발현양상을 면역조직화학적으로 추구하였던 바 다음과 같은 결과를 얻었다. 사이토크롬 P4501A1/2단백질은 약물처치군과 무처치군에서 동일하게 간소엽주변성으로 강하게 유도발현되었다. 이러한 반응양상은 3-MC및 TCDD와 같은 Ah receptor 외인성 조절요인 물질뿐만 아니라 사구체 경변증을 보이는 무처치군에서의 발현은 내인성 요인들에 의해서도 작용한다고 보고 있으며, 특히 신변증의 경우에 나타나는 내인성 조절물질에 의해서 Ah receptor와 관계된 사이토크롬 P4501A1/2단백질 발현기전에 대하여 새로운 관점을 제시하고 있다.
