  • The synthesis and use of cobaltic EDTA as an extracellular space marker has been applied to rat liver for determining and calculating hepatic metabolic changes. The cobaltic form in extracellular space is cobalt-ethylenediaminetetraacetic acid(Co-EDTA). The cobaltic ion has a much higher affinity for EDTA as a potassium salt and is apparently biologically inert. Cobalt-EDTA complex 1.2mM/200g rat body weight was injected initially into rats portal veins to get optimal plasma concentration in rats hepatic veins. Hepatic extracellular water compartments may assume 25% of total hepatic water, indicating Co-EDTA is accessible. Cobaltic for hepatic extracellular accessible was estimated 0.21±0.002㎖ H₂O/g wet weight of freeze clamped liver while 0.19±0.004㎖ of <TEX>$$^{14}C$$</TEX>-inulin was accessible in the our experiment(n=5). Blood plasma water could be calculated as 0.50㎖ water by Co-EDTA and 0.52㎖ by <TEX>$$^{14}C$$</TEX>-inulin by combining the measure of hematocrit from hepatic venous blood. These data well match that of <TEX>$$^{14}C$$</TEX>-inulin experiment. Figure I illustrates the general relationship between plasma and excretion into urine after a single injection through the portal vein in rat liver. It has been noted that plasma levels of Cobalt-EDTA were very low in as determined by furnace methods and is rapidly excreted into urine without any toxicity.
